Messenger RNA (mRNA) quality control (QC) is a critical step in biotechnology and biopharmaceutical manufacturing, ensuring that mRNA molecules used in vaccines, gene therapies, and research applications are intact, correctly synthesized, and free from contaminants. The assessment of mRNA quality involves detecting factors such as sequence fidelity, structural integrity, purity, and stability.
High-quality mRNA must be free from degradation, unwanted modifications (e.g., incomplete 5' capping), truncated transcripts, and contamination from residual plasmid DNA, genomic DNA or proteins. Advanced analytical techniques, including electrophoresis, quantitative PCR, liquid chromatography mass spectrometry (LC-MS), sequencing, and spectroscopic methods, are employed to verify mRNA integrity before use in clinical and research settings.
RNA integrity by Agarose Gel Electrophoresis
RNA integrity by Capillary Electrophoresis
Reverse Transcription Quantitative PCR (RT-qPCR)
Next-Generation Sequencing (NGS) and RNA-Seq
UV-Vis Spectrophotometry (Nanodrop A260/A280 Ratio)
Fluorometric RNA Quantification (Qubit Assay)
mRNA 5′- CAP by RP–LC-MS/MS
Percent POLY(A) TAIL Length by IP-RP-HPLC
dsRNA by IMMUNOBLOT/ELISA
Genomic DNA Contamination Testing via qPCR
Protein Contamination Analysis Using SDS-PAGE or Western Blot
Endotoxin Testing Using Limulus Amebocyte Lysate (LAL) Assay
mRNA quality control is crucial in both research and biopharmaceutical applications, ensuring integrity, purity, and functional efficacy. A combination of electrophoresis, qPCR, sequencing, and biochemical assays provides a comprehensive assessment of mRNA quality. As mRNA-based medicines and vaccines continue to evolve, advanced QC techniques will remain essential in ensuring their safety and effectiveness.
