enhancer trap

An enhancer trap is a molecular biology technique that allows for characterizing detailed gene expression patterns in specific tissues or cells. The technique involves the use of an insertional construct that contains a weak promoter upstream of a reporter gene, such as gusA.

The enhancer trap construct comprises a transposable element, necessary for random insertion into the genome, and a reporter gene, necessary for identifying spatial regulation by the enhancer. When an enhancer regulates gene expression, it also affects the expression of the reporter gene. Thus, expression patterns of the reporter gene can be used as a proxy for the expression patterns of nearby genes at the insertion locus. In enhancer traps, a promoter, usually a P-transposable element, is inserted into the genome together with a reporter gene, such as lacZ and GAL4.
