CHO expression refers to the use of Chinese hamster ovary (CHO) cells as a host system to produce and express recombinant proteins, including therapeutic proteins, enzymes, and other biologically active molecules. CHO cells have become a popular and widely used platform for the production of biotherapeutics and pharmaceuticals due to their ability to efficiently produce complex proteins with proper post-translational modifications. Key features of CHO expression include: 1. High Expression Capacity(CHO): CHO cells have the ability to produce high yields of recombinant proteins, making them suitable for large-scale production. 2. Post-Translational Modifications: CHO cells are capable of performing post-translational modifications, such as glycosylation, which are essential for proper protein folding, stability, and biological activity. These modifications often contribute to the therapeutic efficacy and safety of the final product. 3. Compatibility with Serum-Free Culture: CHO cells can be grown in serum-free or protein-free media, reducing the risk of introducing contaminants and facilitating downstream purification processes. 4. Regulatory Approval: CHO-expressed products have a successful track record of regulatory approval for human use, making them a reliable choice for biopharmaceutical development. The process of CHO expression involves introducing a recombinant DNA construct into CHO cells, typically through techniques like transfection or viral vector-mediated gene delivery. The DNA construct contains the gene of interest (coding for the protein of interest) along with regulatory elements to drive gene expression. Once the gene is introduced, the CHO cells begin to produce the desired protein, which can then be harvested, purified, and formulated for therapeutic use or research purposes.
